U.S. Route 340 (US 340) is a spur route of US 40, and runs from Greenville, Virginia, to Frederick, Maryland.In Virginia, it runs north–south, parallel and east of US 11, from US 11 north of Greenville via Waynesboro, Grottoes, Elkton, Luray, Front Royal, and Berryville to the West Virginia state line.

The Federated Garden Clubs of Maryland declared the U.S. Highway a Blue Star Memorial Highway as a tribute to the United States Armed Forces in 1948, and a 1953 resolution signed by Governor William Preston Lane, Jr., officially dedicated US 301 as such. There are seven mainline sections of MD 144: MD 144 in Allegany County runs 17.55 miles (28.24 km) from MD 807 in Cumberland east to US 40 Scenic east of Flintstone. The state highway generally parallels I-68 and crosses over the freeway multiple times as both highways pass through mountainous creek valleys and over Martin Mountain and Polish Mountain.
The state highway also links Charlottesville to Fredericksburg (via SR 3) by way of Orange County, within which SR 20 is the main east–west highway and intersects US 15 again in the county seat of Orange. SR 20 is a Virginia Byway from Scottsville to Orange.

Bealeton is located in southwestern Fauquier County. U.S. Route 17 leads north 10 miles (16 km) to Warrenton, the county seat, and southeast 27 miles (43 km) to Fredericksburg. VA 28 leads northeast 20 miles (32 km) to Manassas and southwest 2.5 miles (4.0 km) to U.S. Routes 29 and 15, which lead an additional 13 miles (21 km) to Culpeper.
