Polyacrylamide (abbreviated as PAM or pAAM) is a polymer with the formula (-CH 2 CHCONH 2-). It has a linear-chain structure. PAM is highly water-absorbent, forming a soft gel when hydrated. In 2008, an estimated 750,000,000 kg were produced, mainly for water treatment and the paper and mineral industries. [1]

The spacer in the bacterial CRISPR loci will not contain a PAM sequence, and thus will not be cut by the nuclease, but the protospacer in the invading virus or plasmid will contain the PAM sequence, and thus will be cleaved by the Cas9 nuclease. Another advantage of PAMs is their inherent compliant behavior: when a force is exerted on the PAM, it "gives in", without increasing the force in the actuation. This is an important feature when the PAM is used as an actuator in a robot that interacts with a human, or when delicate operations have to be carried out.
