Vitamin D compounds, specifically cholecalciferol (D3) and ergocalciferol (D2) are used in rodenticides due to their ability to induce hypercalcemia, a condition characterized by elevated calcium levels in the blood. This overdose leads to organ failure and is pharmacologically similar to vitamin D's toxic effects in humans.
For example, CYP11A1, an enzyme chiefly known for its role in steroidogenesis, has been found to hydroxylate vitamin D3 at several positions, including C-20, C-22, and C-23, without cleaving the side chain. The resulting metabolites, such as 20-hydroxyvitamin D3 and 20,23-dihydroxyvitamin D3, act as inverse agonists for RORα and RORγ2.
Vitamin D (the inactive version) is mainly from two forms: vitamin D 3 and vitamin D 2.Vitamin D 3, or cholecalciferol, is formed in the skin after exposure to sunlight or ultra violet radiation or from D 3 supplements or fortified food sources.
LD 50 of 4.4 mg/kg has been reported in rabbits, with lethality to almost all rabbits ingesting doses greater than 15 mg/kg. [54] Toxicity has been reported across a wide range of cholecalciferol dosages, with LD 50 as high as 88 mg/kg or LD Lo as low as 2 mg/kg reported for dogs. [55]
The most prevalent side effects include nausea, mild sedation, fatigue, and dizziness. At higher dosages, there is an increased risk for restlessness, insomnia, and tremors. [6] Cariprazine was approved for medical use in the United States in September 2015. [12] It was approved as a generic medication in 2022, [13] but is covered by patents ...
Calcitriol is a hormone and the active form of vitamin D, normally made in the kidney. [8] [9] [10] It is also known as 1,25-dihydroxycholecalciferol.It binds to and activates the vitamin D receptor in the nucleus of the cell, which then increases the expression of many genes. [11]
Alfacalcidol (or 1-hydroxycholecalciferol) is an analogue of vitamin D used for supplementation in humans and as a poultry feed additive.. Alfacalcidol has a weaker impact on calcium metabolism [3] and parathyroid hormone levels [4] than calcitriol; but significant effects on the immune system, including regulatory T cells. [5]
