The hotel's lobby, which connects 43rd and 44th Streets, contains a bar and restaurant. The upper stories originally featured 90 apartments, but these were replaced with 205 guestrooms when Philippe Starck and Gruzen Samton Steinglass Architects converted the Royalton to a boutique hotel in the 1980s.

The 1927 building was designed by architect Andrew J. Thomas, erected as Hotel Duane intended for long-term stays. An article of the era notes, "in design, the exterior of the building indicates a free interpretation of the Italian Romanesque style harmonizing very well with the fraternity clubs building at the north, designed in a similar style."
The hotel has 13 stories and 204 guest rooms. The lobby has a handpainted mural by Damien Gilley. Behind the front desk is a mural by Jason Prouty depicting a scene from brewer Henry Weinhard's Barrel Room. [2] The interior also has "floating shelves, air plants and natural wood features". [3]
