Based on the English Pentonville Prison design model of Joshua Jebb, the site's key feature, the Main Cell Block, was designed by the Comptroller of Convicts Captain Henderson, and completed in 1859. Designed to hold up to 1000 prisoners, it is 145 metres (476 ft) long and four storeys high, the longest and tallest cell range in Australia. [ 1 ]
The prison was built using convict labour in the 1850s. Although a convict-supported settlement was established in Western Australia from 1826 to 1831, direct transportation of convicts did not begin until 1850. It continued until 1868. During that period, 9,668 convicts were transported on 43 convict ships.

The building included hospital wards, a cookhouse and mess shed. Due to overcrowding, a second storey was proposed in 1849, but was not proceeded with. Various extensions were added (a latrine and office block formed infill when built later). The barracks was later used by the industrial school, and by prisoners again in 1888.

The dominant archway of the Prisoners' Barracks extended approximately 10 metres (33 ft) through the building from the Queen Street frontage towards Adelaide Street opening into the large walled yard. The yard was the site of Moreton Bay's first public execution in 1830.
