Mancini was born in Rome, Papal States, and showed precocious ability as an artist.At the age of twelve, he was admitted to the Institute of Fine Arts in Naples, where he studied under Domenico Morelli (1823–1901), a painter of historical scenes who favored dramatic chiaroscuro and vigorous brushwork, and Filippo Palizzi.
In 1844 Francesco Mancini became a student at the Naples Royal Institute of Fine Arts, where he studied drawing and attended Gabriele Smargiassi’s course on landscape from 1846. At the end of the decade he moved on from his academic training and devoted himself to painting from life, during numerous travels in the regions of southern Italy.

He was a member of the French Academy in Rome (1732), the Accademia dei Virtuosi in the Pantheon of Rome (1743 and 1745) and director of the Accademia di San Luca (1750–51).
