The street originally contained a chapel by the quayside, known as St. Mary atte Key. [2] The first recorded reference to Chapel Street dates from 1368 on a deed for a burgage plot. By 1355, the church was deemed too small for Liverpool's growing population and a new church was to be built on land given by the Duke of Lancaster to the burgesses.

In 1207 Liverpool received its charter from King John. By the 1250s a small stone chapel known as St Mary del Quay had been built (the first place of worship on the current site of the church). It probably stood near the site of the present tower, overlooking a quay on the River Mersey. The chapel was used as the main centre of worship until 1355.
The first chapel on the site was built in 1811. The foundation stone was laid in April 1811, and the chapel was opened for worship on 27 May 1812. It cost about £13,000 (equivalent to £1,190,000 in 2023), [2] and provided seating for nearly 2,000 people. The chapel was destroyed by fire on 19 February 1840.

Tithebarn Street is home to several university buildings and along with the surrounding streets is a popular site for student accommodation.
