US 65 enters Missouri between Omaha, Arkansas and Ridgedale, Missouri. The four-lane expressway continues through Hollister and Branson toward the Springfield metro area. Through the Branson area, US 65 is built as a freeway. North of Branson is an interchange with the Ozark Mountain Highroad (a freeway spur) and U.S. Route 160.

A 15- to 20-foot (6.1 m) wide S-shaped bridge spans more than 3,000 ft (910 m) across the Missouri River , connecting Omaha's Riverfront Trail with Playland Park in Council Bluffs.
