The US CDC's COVID-19 laboratory test kit. COVID-19 testing involves analyzing samples to assess the current or past presence of SARS-CoV-2, the virus that cases COVID-19 and is responsible for the COVID-19 pandemic. The two main types of tests detect either the presence of the virus or antibodies produced in response to infection.
On 21 May, researchers at Ben-Gurion University in Israel reported a one-minute coronavirus test with 90% accuracy, based on the "change in the resonance in the THz spectral range" shown by the coronavirus through THz spectroscopy. [91] Nearly two million antibody tests imported into Australia and costing $20 million were declared unusable.

An individual is seropositive, or has seroconverted for COVID-19, once standard techniques are able to detect COVID-19 antibodies in the blood. Seroconversion testing is primarily used to detect individuals who have been infected with COVID-19 in the past who have already resolved their infections.
If a couple is fully vaccinated against COVID-19, why would blood tests show they are negative for COVID-19 antibodies? It probably has to do with the test itself.

[2] [3] The N protein is the most highly expressed of the four major coronavirus structural proteins. [2] In addition to its interactions with RNA, N forms protein-protein interactions with the coronavirus membrane protein (M) during the process of viral assembly. [2] [3] N also has additional functions in manipulating the cell cycle of the ...
