The Mountain Bike Trail is a 6.5-mile trail which circles Stump Pond and is located in the northeastern corner of the park. Some of the other shorter trails are: Clear Lake Trail (3 miles), Nature Trail (0.75 miles), High Lake Trail (1.5 miles), Riverview Trail (2.0 miles), and Brian Plawer Nature Trail (0.9 miles).

The Spanish Moss Trail is a partially-completed, ten-mile multi-purpose rail trail in northern Beaufort County, South Carolina.. Using the former Port Royal Railroad right-of-way now owned by Beaufort-Jasper Water and Sewer Authority, the dedicated trail follows a north–south path from Seabrook to Port Royal while passing through Burton and Beaufort.
In addition to the hiking trails there are a number of horse trails in the southeastern portion of the park and a mountain bike trail at the Anthony Wayne Recreation Area in the northeast of the park. In winter some of the trails are open for cross-country skiing. The hiking trails are maintained by the New York - New Jersey Trail Conference.

The park offers rugged semi-wilderness, rising from sea level to steep coastal mountains of more than 2,600 feet (790 m). The park is a popular spot for running, hiking and horseback riding. Mountain biking is restricted to the fire road as of 2004 because of deed restrictions regarding the state park.
