US 281 has two signed segments near its southern terminus in the Rio Grande Valley, both of which are signed west–east.The eastern segment, considered mainline US 281 by TxDOT, begins in Brownsville at an intersection with Business US 77 and SH 48 about 2 miles (3.2 km) from the Mexico border, and travels west through several communities along the border to Pharr.

The section north of Wichita Falls was cancelled on July 15, 1935, [8] but restored on February 11, 1937. [9] The route was previously proposed as the northern sections of SH 22 and SH 23 . The intersection of SH 79 and SH 25, in Archer City , was the location for the filming of movie The Last Picture Show in 1971.
On November 7, 1958, US 287 was rerouted in downtown Wichita Falls, and the FM 171 designation was extended along Lincoln Street to its current western terminus. The eastern terminus was extended to the south, first 2.5 miles (4.0 km) to a county road on May 2, 1962, and then 4.6 miles (7.4 km) to its current location at FM 2332 on June 1, 1965.
