The church was founded in 1903 as Tuam Avenue Baptist Church. It moved to its current location on South Main Street in 1930 and was renamed South Main Baptist Church. Between 1934 and 1939, the church hosted the University of Houston campus before the university moved to its current location on Cullen Blvd.
The church then purchased a new building, located at 975 North Miami Beach Blvd. for $5.60 million in March 2023, according to Miami-Dade County property records.

[2] On January 24, 2005, a fire destroyed the interior of the church, leaving only the exterior brickwork intact. [3] In 2009, the church was sold to the City of Houston.
