The Shire is governed and administered by the Mount Alexander Shire Council; its seat of local government and administrative centre is located at the council headquarters in Castlemaine. It also has service centres located in Maldon and Newstead. The Shire is named after the first European settlers' name for the Castlemaine region and goldfields.
Mount Alexander Shire Council is composed of eight single-member wards. The council increased from seven members to eight prior to the 2024 election; members were previously elected across five wards (four single-member and one three-member). [24]

In October 1915 Newstead Shire absorbed Mount Alexander United Shire, which had been created in June 1871 by amalgamating Campbell's Creek Road District, Fryers Road District and Guildford Borough. The expanded shire was renamed Newstead & Mount Alexander Shire in May 1916. The name reverted to Newstead Shire in May 1949.
