The Yokosuka-Yokohama section was designated as National Highway 45 on 8 July 1887 and became National Highway 31 in 1920 and Route 16 in 1952. On 1 April 1963, Route 16 was extended to Kisarazu, replacing a portion of Route 127 and all of Route 129 when these sections were promoted to Class 1 highways, forming the current route around Tokyo.

The route is a 19.7-kilometer (12.2 mi) long radial highway running southwest from the southern terminus of the Haneda Route in Ōta near Haneda International Airport in Tokyo to the Kariba Route in Naka-ku, Yokohama in Kanagawa Prefecture. Alongside the Haneda Route, it connects Tokyo's Inner Circular Route in central Tokyo to Yokohama.

The Tokyo Expressway is a 2.0-kilometer-long (1.2 mi) privately-owned highway enclosing all but the eastern border of Tokyo's Ginza district.
