The event is named after Mill Reef, a highly successful racehorse in the early 1970s. He was trained at Kingsclere, located several miles from Newbury. The Mill Reef Stakes replaced a similar race, the Crookham Stakes, in 1972. The winner of the inaugural running, Mon Fils, went on to win the following year's 2,000 Guineas.
Mill Reef was given a rating of 141 by Timeform and is listed at no. 4 in the greatest racehorses of the twentieth century. The Mill Reef Stakes at Newbury is named in his honour. The following is inscribed on the plinth beneath his statue at the National Stud: Swift as a bird I flew down many a course. Princes, Lords, Commoners all sang my praise.

Mon Fils (1970–1979) was a British Thoroughbred racehorse and sire, best known for winning the classic 2000 Guineas in 1973. As a two-year-old, Mon Fils won two of his seven races, including the Mill Reef Stakes.
Known Fact (17 March 1977 – 12 July 2000) was a Kentucky-bred British-trained racehorse and sire. He was the leading British miler of 1980, being awarded the 2000 Guineas on the disqualification of Nureyev and defeating Kris to win the Queen Elizabeth II Stakes.

Ribchester (foaled 25 March 2013) is an Irish-bred, British-trained Thoroughbred racehorse. As a two-year-old in 2015 he finished second in the Gimcrack Stakes on his second racecourse appearance and then won the Group Two Mill Reef Stakes.
Shirley Heights was a bay horse bred by his owners, the father and son team of Charles Wood, 2nd Earl of Halifax, and Lord Irwin.He was one of many successful racehorses sired by Mill Reef, an American-bred horse who won the Epsom Derby, King George VI and Queen Elizabeth Stakes and Prix de l'Arc de Triomphe in 1971.
