The PUP Open University was tasked to administer and maintain the campus and its facilities. PUPQC was formally inaugurated on July 29, 1997. Although it was in existing prior to 1997, it only started its operations and formal classes after its inauguration.
A total of 36,527 students in the Manila campus alone are enrolled in the Baccalaureate Program, while 1,658 undergrads are taking up Diploma Courses. About 897 students are also enrolled in the undergraduate programs of the PUP Open University and ETEEAP/Non-Traditional Programs. PUP operates year-round with two semesters and a summer.

PUP San Pedro Campus was established on April 9, 2002, as an open university center by virtue of a memorandum of agreement signed by then-Mayor Felicismo A. Vierneza of San Pedro, Laguna and then PUP President Ofelia M. Carague. PUP San Pedro initially offered a single curricular program, Bachelor of Science in Entrepreneurial Management.
