Route 116 starts at its western terminus at U.S. Route 59 in Rushville and continues east. It then has a t-intersection with Supplemental Route HH and continues south. It then turns east again after an intersection with Supplemental Route M, before curving to the north. It then heads east again and then intersects with Missouri Route 371.
