Main Street is part of the east–west New York State Route 5 (NY 5). American surveyor Joseph Ellicott had the "Great Iroquois Trail", an ancient path from the Hudson River to Lake Erie, improved. It was known as the "Buffalo Road" and, later, "Main Street".
The Williamsville Christian Church, also known as the Meeting House, is a historic Disciples of Christ church located at Williamsville in Erie County, New York. It is a 2 + 1 ⁄ 2 -story brick Italianate -style structure constructed in 1871 and remodeled in about 1900.

As newlyweds, they moved to a log cabin on a farm on the 4301 Main Street location of Amherst Central High School. In 1985 the Williamsville East Boys' Gymnastics also set another record, 99 wins and 0 losses. East was one of four schools left in New York with a boys' team. Their team finished its 2014 season with a shared sectional title with Williamsville South High School and a 12–0 record. [6]
