US 65 passes through Greenbrier, Clinton, Marshall, and Valley Springs. US 65 will join US 412 and 62 before passing through Harrison. After leaving Harrison, US 412 and 62 separates from US 65 at a freeway-to-freeway interchange. US 65 becomes a divided highway. US 65 continues north to the Missouri state line and enters Missouri.

From 1922 to 1926, US 65 in Missouri was known as Route 3.US 65 originally followed Route 248 and US 160 between Branson and Springfield.Route 3 was originally planned on a shorter route between Springfield and Preston, with Route 71 on the longer alignment via Buffalo, but Route 3 was quickly shifted east, absorbing Route 71.

At milemarker 116, I-65 passes Crown Hill Cemetery, burial site and memorial of former-President Benjamin Harrison. I-65 leaves the I-465 loop on the northwest side of Indianapolis. The highway travels past Eagle Creek Park and it then passes the terminus of I-865 and picks up US 52.
By 1926, NC 65 was rerouted along Fifth Street from Third Street. In 1929, NC 65 was adjusted along a more straight route between Winston-Salem and Stokesdale, bypassing south of Belews Creek. In 1930, NC 65 was extended east through Yanceyville to NC 55/NC 57, in Roxboro. [4] In 1931, NC 65 was decommissioned in favor of an extension of NC 48.
