Mesquite is located in southeastern Doña Ana County in the Mesilla Valley, on the east side of the Rio Grande. Interstate 10 passes just east of the CDP, with access from Exit 151. I-10 leads northwest 13 miles (21 km) to Las Cruces , the county seat , and southeast 35 miles (56 km) to El Paso, Texas .
State Highway 20 (SH 20) is a 78.1-mile (125.7 km) highway maintained by the Texas Department of Transportation (TxDOT) that runs from New Mexico State Road 460 at the state line between Texas and New Mexico at Anthony in El Paso County to Interstate 10 at McNary in Hudspeth County. It largely follows a former alignment of U.S. Route 80.
In October 1846, General Stephen Watts Kearny and his dragoons with their scout Kit Carson found the route over the mountains from the El Camino Real de Tierra Adentro on the Rio Grande, via the Santa Rita mines to the Gila River which he then followed to the Colorado River, at the Yuma Crossing where he crossed the river and then the Colorado Desert to Southern California.
In 1946, US 85 was extended south to the Mexican border in El Paso, multiplexing with US 80 from Las Cruces into Texas.
