Las Vegas was founded as a city in 1905, when 110 acres of land adjacent to the Union Pacific Railroad tracks were auctioned in what would become the downtown area. The first hotel in the area, the Hotel Nevada, was built in 1906; it is still operating as the Golden Gate Hotel and Casino.

The Fremont Hotel and Casino is one of the casinos and hotels currently located in Downtown Las Vegas that is part of the Fremont Street Experience. The casino is located on what is commonly referred to as the four corners. These are the four main hotels that are located on the corner of Casino Center Boulevard and Fremont Street.

A 1952 map of downtown Las Vegas (depicting road layouts as of late-1950) shows the highway alignment with the state route number and without the US highway designation. The route follows Rancho Drive, a major arterial connecting downtown Las Vegas to the northwest part of the city. Much of SR 599 was previously designated as U.S. Route 95 (US 95) prior to completion of the Las Vegas Expressway. The route is also designated as U.S. Route 95 Business (US 95 Bus.).
