Gram stain (Gram staining or Gram's method), is a method of staining used to classify bacterial species into two large groups: gram-positive bacteria and gram-negative bacteria. It may also be used to diagnose a fungal infection . [ 1 ]

In bacteriology, gram-positive bacteria are bacteria that give a positive result in the Gram stain test, which is traditionally used to quickly classify bacteria into two broad categories according to their type of cell wall.
Eubacterium is a genus of Gram-positive bacteria in the family Eubacteriaceae. These bacteria are characterised by a rigid cell wall. They may either be motile or nonmotile. If motile, they have a flagellum. A typical flagellum consists of a basal body, filament, and hook. The long filament is the organ which helps eubacteria move.
The general secretion (Sec) involves secretion of unfolded proteins that first remain inside the cells. In Gram-negative bacteria, the secreted protein is sent to either the inner membrane or the periplasm. But in Gram-positive bacteria, the protein can stay in the cell or is mostly transported out of the bacteria using other secretion systems.
The outer red layer in this diagram is the capsule, which is distinct from the cell envelope. This bacterium is gram-positive, as its cell envelope comprises a single cell membrane (orange) and a thick peptidoglycan-containing cell wall (purple). The bacterial capsule is a large structure common to many bacteria. [1]
