It is often desirable to know the unordered amino acid composition of a protein prior to attempting to find the ordered sequence, as this knowledge can be used to facilitate the discovery of errors in the sequencing process or to distinguish between ambiguous results.
The generated protein is a sequence of amino acids. This sequence is determined by the sequence of nucleotides in the RNA. The nucleotides are considered three at a time. Each such triple results in addition of one specific amino acid to the protein being generated.
Methods for performing protein sequencing include: Edman degradation; Peptide mass fingerprinting; Mass spectrometry; Protease digests; If the gene encoding the protein is known, it is currently much easier to sequence the DNA and infer the protein sequence.
