SR 79 is a south-to-north highway running though eastern Holmes County. The road enters the county from Washington County to the south as passes through Bonifay, Holland Crossroads and Esto, before finally crossing the Florida-Alabama State Line where it turns into AL 167. SR 81 is a south-to-north highway running through western Holmes County.
County Road 277 is a county extension of SR 277 known as Fanning Branch Road. The route begins at Holmes Valley Road which is an unmarked dirt road on both sides of the southern terminus. The next intersection at the corner of a local church is Reno Road, which is paved and also leads to Holmes Valley Road which itself is paved east of that point.
County Road 2A, formerly State Road 2A, exists in two segments. The first is a short spur that runs into the town of New Harmony in Walton County, and the second runs from the Walton-Holmes County line, through Royals Crossroads and SR 2/SR 81, to CR 185. The road is signed as County Road 2A.
