I constructed a much improved Truffle Worm autofarm after the changes to Truffle Worms in 1.3. Truffle Worms can now be killed by hostile mobs, and hostile mobs are much more difficult to kill via traps in expert mode, so the previous version of the farm is no longer very effective.
Truffle worms are created as one possibility when a spawn event lands on mushroom grass. Thus a higher spawn rate, and higher number of spawn events, will make for a higher production rate of Truffle Worms. The is a 1 divided by {spawn rate} chance of a 'spawn event' on each game tick (60 ticks per second, in line with the normal frame rate).
The Truffle Worm farm is also key to the fastsest rate of farming developer armor. Farming the Golem is good too, but the rate of Power Cells per hour pales in comparison to the rate of Worms per hour, and it is very easy to take down Fishron with endgame gear. There are more advanced and efficient designs for Truffle Worm farms out there, but it can require setting up wiring and using traps obtained from the Lihzahrd Temple. Basically I pick out an area of the Cavern layer, clear the blocks out of it, and build a series of Mud Block walkways divided by 3 spaces, so that you can walk in all the sections.

Truffle Worm Autofarm (Ideal): The Truffle Worm autofarm should be built in the jungle biome in the stone layer. With a mounted Water Candle and a fast kill rate, the spawn rate reaches a cap of 60 (which corresponds to 1/60th of a chance to spawn a mob every 1/60th of a second).
According to the Wiki, Truffle Worms are classed as enemies, which means that you can raise their spawn rate through the use of Water Candles and Battle Potions. Pop a few Water Candles down in the area and repeatedly move from one side of the box to the other.
