The 556th Bombardment Squadron was activated at MacDill Field, Florida on 1 December 1942 as one of the four squadrons of the 387th Bombardment Group and trained at bases in the southeastern United States with Martin B-26 Marauder medium bombers until June 1943, when it deployed to the European Theater of Operations.

The 556th Strategic Missile Squadron became completely operational on 15 September 1964. This was the last Atlas squadron to be accepted and the only Intercontinental ballistic missile base east of the Mississippi River. The 556th's last operational day was 30 April 1965 with the squadron's inactivation on 25 June 1965.
