In Houston County, the route has no major junctions until it reaches the Ross Clark Circle, which is US 84 and SR 210. This same junction is the junction with US 231 Bus. and the point where SR 53 ultimately leaves its state-wide concurrency with US 231. The route turns south onto the Ross Clark Circle and heads to the junction with US 84 Bus.
Southern terminus at I-10 in Mobile. I-65 starts its northward journey in Mobile as a three-lane freeway at a directional T interchange with I-10 not far from the Gulf of Mexico. From there, it runs northeast, intersecting with I-165 in Prichard. At exit 13 in Saraland, the Interstate transitions from three to two lanes.
The route then continues south to SR 287, also known as the Gulf Shores Parkway. That route is an effective route for people heading to Montgomery and beyond from Gulf Shores. The routes swap right-of-ways and SR 59 continues along the Gulf Shores Parkway. The route reaches US 31 soon. The two routes engage in a concurrency to Stapleton.
The section of US 90 that is multiplexed with I-10 through Houston is the only section of the route that is unsigned. In eastern Houston, US 90 splits from I-10 and heads northeast towards Liberty , eventually traveling through downtown Beaumont where it rejoins I-10 for the rest of its routing through Texas.
Upon leaving US Route 90 and US Route 98 in Spanish Fort, the route heads due east towards the town of Stapleton, where it joins Alabama State Route 59 (Gulf Shores Parkway). About eleven miles later, the route leaves the Gulf Shores Parkway right-of-way and follows a town square around the Baldwin County Courthouse in Bay Minette.
