In the issue, witches from the DC and Looney Tunes universes placed a wager where the existence of all birds and cats (as well as all bird- and cat-themed heroes and villains) depended on if Sylvester could eat Tweety. Sylvester (designed more realistically for the DC Universe) teamed up with Catwoman, while Tweety teamed up with the Black Canary.
It is the only scene in which Granny appears, as the rest of the cartoon is devoted to Sylvester's latest attempts to catch and eat Tweety, which begins when, after failing to grab a fish by hiding in a fisherman's basket (the fisherman caught a crab instead, which Sylvester gets attacked by), he sees the boat carrying the canary chug past.
Sylvester wants to catch and eat Tweety, but some major obstacle stands in his way – usually Granny or her bulldog Hector (or occasionally, numerous bulldogs, or another cat who also wants to catch and eat Tweety). Tweety says his signature lines "I tawt I taw a puddy tat!" and "I did! I did taw a puddy tat!"

Granny leaves Hector in charge of looking after Tweety while she is away. If he fails to protect Tweety, she will shoot Hector. Sylvester tries to eat Tweety, first by disguising himself as a scarecrow (complete with ragged clothes and tall stilts), then Hector starts barking and an annoyed Sylvester hits him with his wooden leg and yells his only spoken line in the cartoon, "Aaaaah, Shaddap!"
The Sylvester-chases-Tweety cycle is continually disrupted by a mouse who is even hungrier and more desperate than Sylvester, the mouse seizing upon the idea of eating Sylvester. Sylvester suggests that Tweety play sailboat, but in a pot where he is attempting to cook him. This is interrupted by the mouse discovering Sylvester and gnawing his tail.
