Wanneroo Road is part of State Route 60, as an alternative route to the Mitchell Freeway (State Route 2) in Perth's northern suburbs. The road's southern terminus is London Street (State Route 61) in Joondanna, where it continues southwards as Charles Street, and its northern terminus is at Yanchep Beach Road in Yanchep, where it continues northwards as Indian Ocean Drive.

Wanneroo covers a land area totalling 685.8 square kilometres (264.8 sq mi). The city is bounded by Beach Road and the City of Stirling to the south, Alexander Drive and the City of Swan to the east, Wanneroo Road and Lake Joondalup to the south-west, the Indian Ocean to the west and the Shire of Gingin to the north.
Route 468 is a much faster service, passing straight through Wanneroo along Wanneroo Road. All three of these bus services deviate to the Wanneroo Secondary College at school start and end hours. There are no bus services serving the housing areas to the west side of Wanneroo Road.
Neerabup is home to the Wanneroo Raceway, a 2,411-metre (1.498 mi) road racing circuit. Wanneroo Raceway, which opened in 1969, is the home of motor racing in Western Australia and hosts an annual round of the Supercars Championship. Neerabup is also home to the Pinjar Park Speedway which opened in 2005.
Nowergup is a sparsely populated agricultural suburb. Several plant nurseries, the Ocean View Tavern and the Nowergup Lake wetland and fauna sanctuary are situated along Wanneroo Road. The western strip between Wanneroo Road and the Mitchell Freeway is approximately the northern half of the Neerabup National Park.

In 1908, following a request from 50 local residents, the Wanneroo Road Board (which eventually became City of Wanneroo) successfully applied for a 20-hectare (49-acre) reserve for camping and a health resort. By the 1920s, the area was well used by locals, who knew the area as "Burns Beach" after a farmer who ran sheep in the area.
