McGuire Air Force Base was established as Fort Dix Airport in 1937 and first opened to military aircraft on 9 January 1941. On 13 January 1948 the United States Air Force renamed the facility McGuire Air Force Base in honor of Major Thomas Buchanan McGuire Jr., (1920–1945).
The 305th Bomb Group bombed the navy yards at Wilhelmshaven on 27 January 1943 when heavy bombers of Eighth Air Force made their first penetration into Germany. Through mid-1943, the group attacked strategic targets such as submarine pens, docks, harbors, shipyards, motor works, and marshaling yards in France, Germany, and the Low Countries .

In Ju1y-October 1980, it helped airlift an F-4 wing to Egypt in support of first the joint USAF/Egyptian Air Force exercise. On 9–12 May 1983, the first all-woman C-141 crew, members of the 18th MAS, flew from McGuire AFB to Rhein Main AB, Germany on an aeromedical evacuation mission.

The US Air Force still operates a small fleet of drone recovery vessels nicknamed the "Tyndall Navy". These ships recover pieces of wreckage from drones and aerial targets from the waters of the Gulf of Mexico. The largest of these vessels are three 120-foot ships operated by the 82nd Aerial Targets Squadron, which is based at Tyndall AFB, Florida.

The 108th Wing is a unit of the New Jersey Air National Guard, one of the many units stationed at the McGuire Air Force Base entity of Joint Base McGuire-Dix-Lakehurst, New Jersey. If activated to federal service, the Wing is gained by the United States Air Force Air Mobility Command .
