U.S. Route 49 (US 49) is a north–south United States highway. The highway's northern terminus is in Piggott, Arkansas, at an intersection with U.S. Route 62 (US 62). Its southern terminus is in Gulfport, Mississippi, at an intersection with US 90. US 49 is approximately 516 miles (830 km) in length.

U.S. Highway 49 Business (US 49B and Hwy. 49B) is a 3.66-mile (5.89 km) business route of U.S. Route 49 in Craighead County. [5] The route's southern terminus is at US 49 and Highway 1 (AR 1) in south Brookland. It runs north as Holman Street, passing the Brookland City Hall and Brookland High School.

The southern terminus of I-49 is located at a cloverleaf interchange with I-10 and US 167 in the southern Louisiana city of Lafayette.Southbound motorists continue through the interchange onto the Evangeline Thruway, which transitions from a limited-access portion of LA 182 to a major divided thoroughfare that picks up the US 90 corridor heading through the heart of Lafayette.

The highway then narrows to two-lanes to leave Indianola and cross the Sunflower River for a second time. Prior to this point, the entire route of US 49W is a four-lane highway. US 49W passes through Sunflower, where it intersects unsigned MS 810 and rejoins MS 3, before passing through Dwyer and Blaine.
