Here are my ideas: Gent, Brussels, Antwerp, Leuven are very easily reached, 30-50 minutes by direct train, and they are all worth a day. It's always worth seeing The Grand Place and I think easy to fit in your travel plans. It's just needs some 5 to 10 minutes walking from Brussels Central railway station and in case passing through Brussels you hop after visiting it on one of the very frequent later trains and continue with the same ticket.

Mechelen, Antwerp, Rotterdam, Delft, The Hague, Haarlem, and Utrecht can all be more-or-less on your way, depending on how you do it, and are all worth a visit.
Felt Brussels was only worth the time to admire the beauty of the Grand Place. Your half day stop would have been enough for us. We did spend the night traveling between Brugge and Amsterdam. We also enjoyed our time in Trier. Stayed in the local convent both times. A car road race took place on one visit with a parade of all the entrants.
Brussels is the least interesting of the cities, a big, dirty, crowded town. The city center is worth a visit, but beyond the Grand Place is just a warren of tourist and waffle shops. Brussels is worth a day, but I think most visitors are not quite as wowed by it as they are by the alternatives. By all means, spend a day in Brussels (or at least part of a day - you can at least get a look at the top sights in an afternoon).
Daytripping by unreserved local train makes this a trivial outing. Those with lower budgets might stay in Ghent and daytrip to Bruges. Antwerp and Brussels (my least favorite) are also easily accessible. If I had time before a BRU flight, I might visit Leuven as well. Ghent is 30 to 45 minutes from Bruges by train.
