6-MeO-DMT, or 6-methoxy-N,N-dimethyltryptamine, also known as 6-OMe-DMT, is a serotonergic drug of the tryptamine family. [ 1 ] [ 2 ] It is the 6- methoxy derivative of the serotonergic psychedelic N , N -dimethyltryptamine (DMT) and is a positional isomer of the serotonergic psychedelic 5-MeO-DMT .
[5] [22] [1] [4] In fact, there may even be sensitization to the effects of 5-MeO-DMT. [4] The lack of tolerance development with 5-MeO-DMT may be due to biased agonism of the serotonin 5-HT 2A receptor. [5] More specifically, 5-MeO-DMT activates the G q signaling pathway of the serotonin 5-HT 2A receptor with much less potency in recruiting β ...
[164] [166] [167] Similarly to DMT, 5-MeO-DMT is a biased agonist of the serotonin 5-HT 2A receptor, with minimal β-arrestin2 recruitment, and likewise has been associated with little tolerance to its hallucinogenic effects. [168] [142] As DMT has been shown to have slightly better efficacy (EC 50) at human serotonin 2C receptor than at the 2A ...
It is the 5-methyl thio analog of dimethyltryptamine (DMT). 5-MeS-DMT was first synthesized by Alexander Shulgin. In his book TiHKAL (Tryptamines I Have Known and Loved), the minimum dosage is listed as 15-30 mg. [1] The duration listed as very short (less than one hour), just like DMT. 5-MeS-DMT produces similar effects to DMT, but weaker.
At 100 mg, effects began within 5 minutes, peaked at 35–40 minutes, and lasted up to 90 minutes. Higher doses produced effects that were described as psychedelic, such as "swirling, colored patterns typical of tryptamines, tending toward the arabesque". Free base bufotenin taken sublingually was found to be identical to intranasal use.

[1] α,N-DMT is known to be a potent monoamine oxidase inhibitor and tryptamine or serotonin receptor antagonist. [2] [1] Close analogues of α,N-DMT, such as α-methyltryptamine (αMT), are known to act as monoamine releasing agents and serotonin receptor agonists. [3] α,N-DMT is the N-methylated analogue of αMT. Fragment of a hypodermic needle stuck inside the arm of an IV drug user (x-ray). Drug injection is a method of introducing a drug into the bloodstream via a hollow hypodermic needle, which is pierced through the skin into the body (usually intravenously, but also at an intramuscular or subcutaneous, location).
